Sorghum Agar Exploration – part 3


Happy Sunday to all, just continuing to share some progress data so far on the sorghum agar using recipes and also comparing to LME and here are some photos for comparison with few cultures and my preliminary assessment.


Please note the lime green is LME, the lighter shade green is 8gs sorghum with yeast, the darker blueish one is 15g sorghum and the golden one is 15g sorghum with yeast (see full recipes below, all for 500ml water):


  • LME: 7g lme 0.5g yeast extract

  • Sorghum light green – 8g sorghum syrup, 0.5gs yeast extract

  • Sorghum dark blueish – 15gs sorghum (no yeast)

  • Sorghum Golden – 15gs sorghum and 0.5g yeast extract.


Observations:

Seems more is better with sorghum syrup. The 8g recipe is definitely way too weak for most cultures to thrive and the growth is extremely wispy, almost like water agar. This is an easy one to discard so wont be doing this recipe again.


While the 15g sorghum without yeast did better than the 8g, most cultures still were having wispy kind of growth, spotty growth like lacking vigor


The 15g sorghum with yeast did the best in all cultures , showing much stronger and vigorous growth which is what we want here.


Comparing 15g sorghum with yeast with some of the tomentose cultures on LME as shown in the pic, was able to convert them into rhizomorphic growth cases (yes i have tried extremely low lme in those before and still had that growth)


The higher amount needed pf sorghum syrup vs LME makes sense since LME is malt Extract ( no water) vs sorghum syrup has lots of water content due to being a syrup. Im assuming if were using light malt syrup, we would be doing the same as well.


Considering the 15gs sorghum with 0.5gs yeast did the best, i will an even higher nutrition recipe on next go to see how that compares with this one ( my new base). I might try 20gs with 0.5g yeast and/or 15gs sorghum with 1g yeast.
